Acetylcholinesterase

HYB 190-01-02 is specific for brain AChE and does not recognize AChE from erythrocytes. The antibody can thus distinguish between mammalian brain AChE and eruthrocyte AChE. Weak cross-reactivity with Torpedo

marmorata AChE but none with AChE from electric eel or human BtChE.

HYB 190-01-02 has been successfully used in ELISA and Western blotting techniques. Can be used in ELISA on amniotic fluid for the diagnosis of neural tube defects. HYB 190-01-02 is well suited as catching antibody (on an anti-mouse IgG coat) in enzyme antigen immunoassay (EAIA), where the antigen (AChE) is captured and used directly as substrate for acetylthiocholiniodide (Ellmann’s reaction). In Western blotting and dot blotting HYB 190-01-02 reacts with native and denatured human and bovine, detergent soluble and salt-soluble AChE. No cross-reactivity is seen with erythrocyte AChE.

The HYB 190-01-02 immunogen is the C-terminal 10 residues of brain acetylcholinesterase (human and bovine), absent from the eruthrocyte enzyme. Coupled to PPD with glutaraldehyde.

The epitope of HYB 190-01-02 is C-terminal 10 residues (aa 574-583) of brain acetylcholinesterase (DS-AChE and SS-AChE).

Acetylcholinesterase (AChE) is an enzyme located in the postsynaptic membrane and in the muscle endplates, where it hydrolyses the neurotransmitter acetylcholin. AChE from brain is a tetramer (G4-AChE) with a molecular mass of 320 kDa, AChE from erythrocytes is a dimer (G2-AChE) with a molecular mass of 170 kDa. Detection of higher levels of AChE in amniotic fluid can indicate fetal malformations such as neural tube defects.

E-Cadherin

MA1-37202 detects Cadherin E from human samples.

MA1-37202 has been successfully used in immunohistochemistry (paraffin) applications.

The MA1-37202 immunogen is a recombinant protein containing human Cadherin-E residues 600-707.

Cadherins constitute a family of transmembrane glycoproteins involved in Ca2+-dependent cell-cell interactions. The members of this family are differentially expressed in various tissues. They function in the maintenance of tissue integrity and morphogenesis. Cadherins are divided into type I and type II subgroups. Type I cadherins include epithelial cadherin (E-cadherin, cadherin-1 or uvomorulin), neural cadherin (N-cadherin or cadherin-2), placental cadherin (P-cadherin or cadherin-3) and retinal cadherin (R-cadherin or cadherin-4), whereas kidney cadherin (K-cadherin or cadherin-6) and osteoblast cadherin (OB-cadherin or cadherin-11) are type II cadherins. One of the best characterized cadherins is E-cadherin, a ~120 kD transmembrane glycoprotein consisting of an ~80 kD extracellular and a ~40 kD transmembrane and cytoplasmic part. The extracellular domains of E-cadherin are responsible for calcium binding which allows for homophilic interaction with other E-cadherin molecules on the same cell and neighbouring cells. In addition, E-cadherin can interact heterophilically with integrin alphaE-beta7. The cytoplasmic domain of E-cadherin is linked to the actin cytoskeleton through the associated cytoplasmic catenin proteins, thus establishing a complex localized to adherens junctions. In carcinomas E-cadherin is frequently downregulated, which is consistent with its function of an invasion suppressor in normal epithelia.

KELL

MA1-25131 detects KELL from human samples.

MA1-25131 has been successfully used in immunoprecipitation procedures.

Macrophage

MA1-38068 detects Macrophage from human, mouse, rat, and porcine samples.

MA1-38068 has been successfully used in immunohistochemistry (paraffin) applications.

OATP2

MA3-934 detects OATP from human tissue and cells. This antibody does not detect dog, mouse or rat protein. MA3-934 shows no cross-reactivity to other OATP isoforms.

MA3-934 has been successfully used in immunofluorescence and immunoprecipitation procedures. Immunofluorescence staining of OATP2 in human liver samples results in a pattern consistent with plasma membrane staining.

The MA3-934 immunogen is a synthetic peptide corresponding to residues E(671) S L N K N K H F V P S A G A D S E T H C(691) of human OATP2.

The OATPs (organic anion transporter polypeptides) are a family of transport proteins that have been found to mediate the uptake of organic ions into hepatocyte cells. These proteins fall into the solute carrier family 21A (SLC21A), and are expressed in liver, kidney, and brain. These polypeptides are involved in the excretion of potentially toxic compounds and therefore play a role in the overall detoxification system of the body. Unlike other members of the OATP family, OATP2 and 8 have been detected exclusively in the liver, and there is an 80% amino acid sequence homology between these polypeptides. Other names for OATP2 include SLC21A6, OATP-C, and LST-1.

Rad51

MA1-38528 detects Rad51 from human, mouse, and rat samples.

MA1-38528 has been successfully used in Western blot, immunoprecipitation, and immunohistochemistry (paraffin) applications.

The MA1-38528 immunogen is recombinant Rad51 protein.

Rad51 (a eukaryotic homologue of Escherichia coli RecA) is similar to RecA, both biochemically and structurally: It promotes homologous pairing and strand exchange within a regular nucleoprotein filament. In yeast, the Rad51 family of related proteins also includes Rad55, Rad57 and Dmc1. In mammalian cells, seven genes in this family have been identified (HsRAD51, XRCC2, XRCC3, RAD51B/hREC2 and HsDMC1, RAD51C, and RAD 51D). Recent studies suggested that Rad51 is involved not only in homologous recombination but also in cell proliferation regulation.

Ubiquitin

MA1-10035 detects Ubiquitin from human, mouse, rat samples.

MA1-10035 has been successfully used in immunofluorescence, immunohistochemistry, Western blot, ELISA, and immunoprecipitation applications.

The MA1-10035 immunogen is ubiquitin, purified from bovine erythrocytes, cross linked with glutaraldehyde

Ubiquitin is a highly conserved globular 76 amino acid protein of about 8.5 kDa molecular weight. It has a important role in the targeting of proteins for proteolytic degradation. Proteins to be degraded are covalantly coupled to the C-terminus of ubiquitin by means of ubiquitin ligases. The ubiquitin itself is frequently also ubiquitinated, producing a polyubiquitin chain.

Urokinase

MA1-35845 detects Urokinase in human samples.

MA1-35845 has been successfully used in immunohistochemistry (frozen), ELISA, western blotting and radioimmunoassay applications.

The MA1-35845 immunogen is from human urinary urokinase.

Urokinase (UK) is a serine protease, which is one of biological plasminogen activators. It is involved in a number of biological functions including fibrinolysis, embryogenesis, cell migration, tissue remodeling, ovulation, and wound healing. It can be obtained from human urine or kidney cell culture.UK is a two-chain glycoprotein containing 411 amino acids with 12 dissulfide bonds. Its molecular weight is 54,000.

Yersinia pestis

MA1-20815 detects Yersinia pestis in bacteria samples.

MA1-20815 has been successfully used in ELISA, immunofluorescence and immunohistochemistry procedures.

The MA1-20815 immunogen is a Yersinia pestis cell preparation.

Y.pestis has a number of virulence factors that enable it to survive in humans by facilitating use of host nutrients, causing damage to host cells, and subverting phagocytosis and other host defense mechanisms. Capsular F1 antigen is a main immunochemical component of Y.pestis surface. F1 subunit of the antigen is originally synthesized from 170 amino acids and has a molecular mass 17.6 K, then, after the elimination of the signal peptide during the secretion onto the surface, a protein with molecular mass 15.6 K and isoelectric point 4.1 is formed.

a1-Antitrypsin

HYB 185-02-02 has specificity for native human a-1 AT.
HYB 185-02-02 has been successfully used in Western blotting and ELISA procedures. HYB 185-02-02 reacts strongly with native a-1 AT. Strong reaction is seen in ELISA with native a-1 AT directly coated onto the microtiter well. In Western blotting after SDS-PAGE, HYB 185-02-02 reacts with native a-1 AT in reduced as well as non-reduced form.
The HYB 185-02-02 immunogen is a-1 AT isolated from human plasma.
The HYB 185-02-02 epitope differs from that of HYB 191-01-02.
a1 -antitrypsin (a-1 AT) is a single polypeptide chain, a typical member of the serine family (serine protease inhibitors), hibiting the enzyme trypsin. a-1 AT is a glycoprotein synthesized in the liver, and is genetically polymorphic due to the presence of over 20 alleles; individuals appropriately homozzygous are deficient in a-1 trypsin and are predisposed to pulmonary emphysema and juvenile hepatic cirrhosis because of alterations in the amino acid and sialic acid components of the glycoprotein. The molecular mass of a-1 AT is ~51 kDa.
